A study published in the journal Nature Human Behaviour analyzed 2,920 proteins in the blood of 42,062 participants from the UK Biobank and found biological signatures associated with loneliness and social isolation. The researchers identified proteins linked to inflammation, antiviral response, and complement system, as well as future connections with cardiovascular diseases, type 2 diabetes, stroke, dementia, and mortality over nearly 14 years of follow-up.

The research used data from 42,062 participants from the UK Biobank and examined 2,920 plasma proteins, looking for proteomic signatures associated with social isolation and loneliness. The technical summary of the UK Biobank reports that these proteins were linked to inflammation, antiviral responses, and complement systems.

The finding does not mean that every lonely person will automatically have a disease. The central point is that the researchers found measurable biological pathways that can help explain why loneliness and isolation appear associated with a higher risk of physical problems and mortality.

Study analyzed proteins in the blood of 42 thousand people

The research was published on January 3, 2025 in Nature Human Behaviour, with authors affiliated with institutions such as the University of Warwick, University of Cambridge, and Fudan University, according to the UK Biobank record.

The scientists analyzed proteomic data from participants of the UK Biobank, one of the largest biomedical databases in the world. The aim was to look for differences in circulating proteins in the blood of people classified as socially isolated or who reported loneliness.

Proteomics allows the observation of thousands of proteins at the same time. Since proteins are involved in inflammation, immunity, metabolism, coagulation, and cell communication, they function as a kind of molecular panel of the organism’s state.

Loneliness and isolation are not exactly the same thing

The study separates two concepts that are often mixed. Social isolation is a more objective condition, linked to the number of contacts, social interaction, and participation.

Loneliness, on the other hand, is a subjective experience. A person can be surrounded by people and still feel lonely, or live with few contacts and not report suffering because of it.

This distinction is important because the two factors can affect the body through partially different pathways. The research investigated precisely which proteins appeared associated with each of these phenomena.

Proteins appeared linked to inflammation and immune response

According to the UK Biobank summary, the proteins associated with social isolation and loneliness were implicated in inflammation, antiviral responses, and complement systems, important components of immune defense.

This suggests that social connection may be linked to deep physiological processes, not just mood, behavior, or routine. The body seems to register part of this state through changes in circulating molecules.

Low-grade chronic inflammation is already being investigated in various modern diseases. Therefore, finding proteins linked to these processes helps bring loneliness closer to more concrete biological mechanisms.

More than half of the proteins were linked to future diseases

The strongest result appeared in the long-term follow-up. The UK Biobank summary reports that more than half of the identified proteins were prospectively associated with cardiovascular disease, type 2 diabetes, stroke and mortality during a 14-year follow-up.

In the article indexed in PubMed, the researchers report a median follow-up of 13.7 years. During this period, 2,695 cases of cardiovascular disease, 892 of dementia, 1,703 of type 2 diabetes, 983 of stroke, and 4,255 deaths were recorded among the participants followed.

These numbers do not prove that loneliness alone caused these diseases. They show a robust association between protein signature, social condition, and health outcomes, requiring caution and scientific interpretation.

Five proteins caught the researchers’ attention

The Mendelian randomization analysis pointed to a potentially causal relationship of loneliness with five proteins: GFRA1, ADM, FABP4, TNFRSF10A, and ASGR1, according to the UK Biobank summary and the Nature Human Behaviour article.

Among them, ADM and ASGR1 received additional support through genetic colocalization, which strengthens the hypothesis of a biological link. The research also associated these proteins with other blood biomarkers and volumes of brain regions involved in emotional and social processes.

This is one of the most important points of the study. Loneliness did not appear only as a psychological report but as a variable connected to molecules that may participate in pathways related to the heart, metabolism, brain, and risk of death.

ADM appeared as an important clue

The protein ADM, or adrenomedullin, was one of the molecules highlighted by the analysis. It appears in the study among the proteins with the strongest genetic evidence in the link between loneliness and health.

This protein is already being investigated in cardiovascular processes, physiological stress response, and vascular regulation. In the context of the study, it appears as a possible partial mediator between loneliness and some health outcomes.

The research does not say that measuring ADM in a common exam would solve the problem of loneliness. What it shows is that specific proteins can help reveal how social experiences connect to bodily processes.

ASGR1 was also associated with cardiovascular risks

The protein ASGR1 also appears among the five proteins pointed out by Mendelian randomization and was one of the two with evidence reinforced by colocalization, along with ADM.

The study states that these proteins identified by Mendelian randomization exhibited broad associations with other blood biomarkers and brain regions linked to emotional, social, and interoception processes.

Interoception is the ability to perceive internal body signals, such as heartbeat, breathing, tension, and discomfort. This helps connect the study not only to the cardiovascular system but also to how the brain monitors the body itself.

Brain, blood, and social life appear connected

One of the most surprising parts of the study is the link between proteins associated with loneliness and volumes of brain regions involved in social and emotional processes. The UK Biobank summary highlights this connection by describing the proteins identified by genetic analysis.

This reinforces the idea that loneliness is not just “being without company.” It involves social perception, emotional interpretation, stress response, behavior, and physical body signals.

The research does not turn loneliness into a blood diagnosis. But it shows that social experiences can have detectable reflections in complex biological systems.

WHO treats loneliness as a public health issue

The World Health Organization states that social isolation and loneliness are increasingly recognized as a public health problem and a public policy issue across different age groups.

The WHO also reports that about 1 in 6 people worldwide reports loneliness, and among the elderly, approximately 1 in 4 lives socially isolated.

This context helps explain why studies like the one from Nature Human Behaviour have gained relevance. Loneliness is not just an individual issue; it has become a collective problem linked to aging, urbanization, work, technology, and mental health.

Study does not prove that loneliness causes all diseases

Despite the strong results, interpretation needs to be careful. The research itself combines observational analyses, longitudinal follow-up, and genetic methods to investigate possible causal pathways, but it does not claim that loneliness alone explains complex diseases.

Cardiovascular diseases, diabetes, stroke, dementia, and mortality involve many factors at the same time. Genetics, diet, sleep, physical activity, income, access to healthcare, smoking, alcohol, and family history also interfere.

The study contributes by showing that loneliness can be part of this set of risks. It does not replace other factors, but it can act as a relevant piece within a larger public health framework.

Result changes the way we look at social relationships

The practical message of the study is strong: social connection should not be seen only as emotional comfort. It may be related to biological processes that influence physical health, aging, and disease risk.

The research published in Nature Human Behaviour showed that proteins associated with loneliness and social isolation were also connected to cardiovascular diseases, type 2 diabetes, stroke, and mortality during the follow-up.

This places social relationships, coexistence, community support, and human bonds at the center of a broader medical discussion. Instead of treating loneliness as a detail of modern life, science begins to treat it as a measurable risk signal.

Loneliness may enter the prevention radar

The study does not offer a simple solution, but points a way: health does not depend only on medication, diet, and exercise. The quality of relationships can also affect relevant biological markers.

Community programs, care for the elderly, support for teenagers, social spaces, psychological counseling, and support networks can play a preventive role when they reduce isolation and persistent loneliness.

The safest conclusion is that loneliness should not be minimized. The blood of thousands of people showed signs compatible with an increasingly evident reality: social bonds are also part of health.
